Research Overview

What Melanotan 2 is

Melanotan 2 (MT-2) is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone. Its structure, Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, contains a lactam bridge between the aspartate and lysine side chains that constrains the peptide into a ring. This cyclisation gives it considerably greater stability and potency than the linear parent hormone. Its molecular weight is approximately 1,024 Da.

Research background

MT-2 was developed at the University of Arizona in the same programme that produced Melanotan 1, and the two are often studied together to compare linear and cyclic analogue behaviour. Unlike MT-1, it is a non-selective agonist with activity at MC1R, MC3R, MC4R and MC5R, which has made it a useful probe in receptor-subtype studies and in animal models of melanocortin signalling. It is also the parent compound of PT-141, which is its C-terminal carboxylic-acid metabolite. Megalab supplies MT-2 for in-vitro research only.

Specification

Each vial contains 10mg of Melanotan 2 as a lyophilised powder. Purity is verified by HPLC at ≥99% on every batch. The current lot is shown in the batch record on this page where published; a Certificate of Analysis is available on request.

Handling and storage

Store unopened vials at −20°C, protected from light, and protect reconstituted solution from light as well. Bring to room temperature before opening. Reconstitute with bacteriostatic water added slowly down the vial wall, swirling gently. Store at 2–8°C after reconstitution and use within 30 days.

Supplied for laboratory research use only.

Reconstitution & storage

Diluent
Bacteriostatic water, added slowly down the inside wall of the vial; swirl, do not shake.
Lyophilised
Store at −20°C, away from light and moisture.
Reconstituted
Refrigerate at 2–8°C.

Frequently Asked Questions

Why is Melanotan 2 cyclic?

A lactam bridge between the Asp and Lys side chains closes the peptide into a ring. The constrained conformation resists enzymatic degradation and improves receptor binding compared with the linear hormone.

How does MT-2 relate to PT-141?

PT-141 (bremelanotide) is the C-terminal acid form of MT-2; the terminal amide is replaced by a carboxylic acid. It was first identified as a metabolite of MT-2.

Which receptors does MT-2 act on?

It is a non-selective melanocortin agonist with activity at MC1R, MC3R, MC4R and MC5R, in contrast to the MC1R-leaning profile of Melanotan 1.
