IGF-1 LR3 (Long R3 Insulin-like Growth Factor-1) is an 83-residue recombinant analogue of human IGF-1. It differs from the native 70-residue protein in two ways: an arginine replaces glutamate at position 3, and a 13-residue extension is added at the N-terminus. Its molecular weight is approximately 9.1 kDa. These modifications substantially reduce its affinity for IGF-binding proteins, which in native IGF-1 sequester most of the circulating molecule.
IGF-1 LR3 is a well-established cell-culture reagent. It is widely used as a mitogenic supplement in serum-free and low-serum media, particularly in the culture of CHO and other mammalian cell lines used for recombinant protein production, where its reduced binding-protein interaction provides more predictable activity than native IGF-1. It is also used in signalling studies of the IGF-1 receptor pathway. Megalab supplies it for laboratory use only.
Each vial contains 1mg of IGF-1 LR3 as a lyophilised powder. Because the compound is a protein rather than a short peptide, the fill weight is lower than other products in the range while still providing ample material for typical cell-culture concentrations. Purity is verified by HPLC at ≥99% on every batch; a Certificate of Analysis is available on request.
Store unopened vials at −20°C, protected from light. Allow to reach room temperature before opening. Reconstitute with bacteriostatic water or sterile buffer added gently down the vial wall; do not vortex, as proteins of this size can denature under shear. Aliquot the reconstituted solution to avoid repeated freeze–thaw cycles and store at 2–8°C for short-term use, within 30 days.
Supplied for laboratory research use only.
Long R3: “Long” refers to the 13-residue N-terminal extension and “R3” to the arginine substitution at position 3. Together they reduce binding to IGF-binding proteins.
IGF-1 LR3 is a 9.1 kDa protein used at low concentrations in cell culture. One milligram provides sufficient material for a large number of typical culture applications.
No. Vortexing can denature proteins of this size. Add diluent gently and swirl, or allow the powder to dissolve without agitation.
